Rapamycin forms a complex with FKBP12 that directly inhibits mTORC1, a master regulator of cell growth and proliferation. This blocks the G1 to S phase transition, controlling cellular expansion and metabolism.

Suppresses Protein Synthesis
By inhibiting mTORC1, rapamycin blocks the phosphorylation of S6K1 and 4E-BP1, critical regulators of protein translation. This reduces synthesis of proteins required for cell growth, slowing cellular machinery that drives proliferation.

Activates Cellular Autophagy
Rapamycin promotes autophagy, a cellular recycling process where damaged proteins and organelles are broken down and removed. This housekeeping mechanism helps maintain cellular quality and extends cellular lifespan.

Modulates Immune Cell Activity
